Laboratory experiments, Aberdeen, Idaho. Data recorded for beta-glucan, protein, and lipid contents. Moisture was determined according to an official method (AOAC 2002) and was used to convert the contents of other components into a dry matter basis. The total nitrogen/protein content was measured by a combustion method (AOAC 2002), using a protein analyzer (Model FP-528, Leco Corp. St. Joseph, MI). The protein content was calculated with a conversion factor of 5.75. The oil (lipid) content was determined by an AOCS Official procedure (AOCS 2005), using a fat analyzer (Model XT 10, Ankom Technology, Macedon, NY). However, instead of using petroleum ether, hexane was used as the extracting solvent. Beta-glucan determinations were based on the method of Hu and Burton, 2008.

Plots were inoculated by spreading infected straw at the base of plants at the mid-tillering stage of growth. Infected straw was collected from heavily infected barley plants grown during the previous season (2003) at the same site. Ratings were made on the percentage of leaf area affected by disease on a 0-100% rating scale and was converted to the 1-9 scale. The average severity disease severity on M61 (susceptible MN breeding line), Stander (susceptible MN cultivar), and resistant control (line NDB112), was 74% (GRIN Score=7), 75% (GRIN Score=7), and 1% (GRIN Score=1), respectively. The spreader in the nursery, Max, became infected later than many entries in the nursery. By the time of the 2nd evaluation, it ranged from 30-80% severity (many of the lines that were evaluated reached 100% severity). Cool conditions were favorable for continued development of stripe rust through early May, though many barley lines in the nursery were becoming mature and tissue was naturally senescing by that time.
