The spring growth habit in wild type Kronos is determined by the dominant Vrn-A1 allele, which is epistatic to the functional vrn-B1 allele for winter growth habit. In the absence of a functional Vrn-A1 locus the plant requires vernalization for early flowering. The vrn1- null mutant flowers very late (Chen and Dubcovsky, 2012). The Kronos vrn1-null mutant combines mutants vrn-A1 (Kronos T4-2268) and vrn-B1 (Kronos T4-2619). The vrn- A1 allele has a splice site mutation at the end of intron 2 that resulted in a 1-bp frame shift mutation and a premature stop codon. The vrn-B1 allele has a splice site mutation that generates an immediate stop codon after the first exon (Chen and Dubcovsky, 2012). To accelerate flowering, the vrn1-null plants were crossed twice with the Kronos vrn2-null mutant developed before (Distelfeld et al. 2009). The selected plants deposited here are homozygous for loss-of-function alleles in both VRN1 and VRN2 homeologs (vrn1 vrn2-null) and head in approximately 80 days without vernalization. A derived line carrying loss-of-function mutations in both VRN2 homeologs was described before (Distelfeld et al. 2009), and was used as recurrent parent. Both Kronos and Krons-vrn2-null have functional copies of the FUL-A2 and FUL-B2 genes. The ful2-null mutant combines mutations ful-A2 (Kronos T4-837) and ful-B2 (Kronos T4-2911). The ful-A2 mutant has a mutation in the splice donor site of the fifth intron. RT-PCR and sequencing analysis of the ful-A2 transcripts revealed two incorrect splicing forms one with a deletion of 14 amino acids in the middle of the K-box (from position 144 to 157) and the other with a premature stop codon. The ful-B2 mutant (Kronos T4-2911) carries a premature stop codon at position 162 (Q162*) that removed the last 13 amino acids of the K-box and the entire Cterminus. The ful2-null mutant shows a significant increase in the number of spikelets per spike and heads at the same time as Kronos-vrn2-null. The FUL-A3 mutant carries a mutation in the splice acceptor site of the third intron, which generates a new splice acceptor site, a shift in the reading frame and a premature stop codon. The FUL-B3 mutant carries a premature stop codon at amino acid position 132 (Q132*) that removes half of the K-box and the complete C-terminus. The ful3-null mutant did not show any evident phenotype when present alone. When ful3-null was combined with vrn1-null and ful2-null, the triple mutant showed reduced stem length and increases vegetative characteristics of the tiller-like lateral organs in the spike (Li et al. 2019).2116455PI 698815
