Langdon [LDN] and RSL65 (Joppa et al. 1997), a near-isogenic recombinant inbred chromosome line of LDN with a 30-cM segment of chromosome arm 6BS from T. turgidum L. ssp. dicoccoides (accession FA15-3, hereafter referred to as DIC; Avivi 1978). This segment from DIC contains two tightly-linked genes of agronomic interest: The high temperature adult plant stripe rust resistance gene Yr36 (Fu et al. 2008) and the high grain protein content gene GPC-B1 (Uauy et al. 2006). To break the linkage between these genes so that they may be manipulated independently in research and breeding programs, a total of 4,500 F2 plants from the cross LDN ? RSL65 were screened for recombination between PCR markers Xucw110 and Xucw125; only twelve F2 recombinant lines (0.3%) were found. Selected plants from five of these lines were self-pollinated and F3 lines homozygous for the critical recombinations were obtained. The 300 g of seed of each line here provided were harvested from the resultant F4 plants. Two of the lines (IR14-40 and 524) carry the resistant allele of Yr36 combined with the non-functional allele of GPC-B1. The other three lines (IR51-8, IR17-47, and 504) carry the high protein allele of GPC-B1 in combination with the susceptible allele of Yr36.
