This gene was likely originated in the European Spelt wheats (Dang et al. 2022). Using exome capture data, we identified five major haplotypes in the Yr78 candidate region, with the H1 haplotype associated with the resistance allele. The H1 haplotype was not detected in tetraploid wheat (Triticum turgidum), which indicated that Yr78 can be used to improve resistance to stripe rust in most durum wheat cultivars. Yr78 was found in ∼30% of the common wheat cultivars (Triticum aestivum), suggesting favorable selection for the H1 haplotype (Dang et al. 2022). To facilitate the deployment of Yr78 in durum wheat breeding programs, we generated the Kronos-Yr78 germplasm, where we transferred Yr78 from hexaploid wheat into the modern high-quality durum wheat cultivar Kronos. Kronos exhibits partial resistance to Pst as a result of three minor partial resistance QTLs, and the addition of Yr78 significantly increased its resistance to Pst in the field in 2023 and in controlled chamber inoculations. Diagnostic molecular markers CDM158 and CDM160 for the H1 haplotype are available to accelerate the deployment of Yr78 in durum or common wheat (Dang, et al. 2022). 2158168PI 702944
