These premature stop codons eliminate the last 241 (A-genome) and 244 (B-genome) amino acids of the C-terminal region, including the two conserved blocks of the ELF3 protein (Alvarez et al., 2016).
For the phyB loss-of-function mutant, we combined the A-genome mutation R586* found in Kronos-2711 with the B-genome mutation Q1027* found in Kronos-2078. These premature stop codons eliminate the last 641 (A-genome) and 144 (B-genome) amino acids of the C-terminal region, so both proteins lack the histidine kinase domain and are likely not functional (Pearce et al. 2016).
The resulting BC2F3 loss-of-function elf3 and phyB lines were intercrossed and lines homozygous for the four mutations (elf-A3 elf-B3 phyB-A phyB-B) were selected with molecular markers and are the material deposited here. These plants head in ~50 days from sowing.
