For each accession six eight-seed samples were crushed and each stained with 200 ul of iodine stain (Carmel = Waxy ; Purple = Nonwaxy). The stock iodine solution had 2mg Potassium Iodide / ml. of water ); the working solution was more dilute, 50 ml of the stock solution mixed with 950 ml of nanopure water. The working solution must be used fresh. A stereo dissecting scope and added light enhanced observations. Since seeds were bulked; lines that were mixed for the waxy trait might not be detected.
