Narrative
Improvement of cold tolerance at key stages of growth and deployment of favorable alleles for root traits and cold tolerance into elite sorghum genetic resources are recognized as critical factors for sorghum production in drier and cooler regions of the US, permitting farmers to capitalize on early season moisture, efficient integrated pest management and to cultivate fuller season varieties with higher yield potential. The BTx623NSZ _ RIPG mapping population consists of 292 inbred lines, was evaluated for expression of early season cold tolerance based on field and controlled condition (15°C) germinability/emergence and seedling vigor; data for these traits are provided from two years of early season field screen and minimum of three laboratory studies. A detailed analysis of gene expression pattern and root morphological traits under cold stress (15°C) were characterized for the parents of the inbred population. Seeds were bulked from 10 representative uniform plants at F6 generation and advanced by selfing to F1 generation. Briefly, BTx623NSZ_RIPG, was developed by hand emasculation of the female parent BTx623 and pollination with PI 568016 in the polyhouse at Cropping Systems Research Laboratory, Lubbock, TX in 2012. The resulting F1 hybrid was intermediate in height between the 2 parents, with brown seed color. A total of 300 F2 plants were planted at the F3 generation followed by careful advancement to F4 to F7 generations performed alternately in Lubbock, Texas and Lajas, Puerto Rico. At the F6:7 stage of development ten representative uniform plants were tagged and seeds were bulked from the 10 tagged plants to compose each line. A total of 292 RILs were generated and are used to represent the BTx623NSZ_RIPG. The population was developed thru cooperative efforts of PSGD team of: Dr. Gloria Burow, Dr. John Burke, Dr. Ratan Chopra, Ms. Halee Hughes, and Mr. Jacob Sanchez.

BTx623 (USDA-ARS GRIN PI 564163), is an elite maintainer line with a pedigree of BTx3197xSCI 70-6-4. PI568016, known as Niu Sheng Zui (NSZ) is a landrace from China classified under nervosum-broomcom working group of sorghum. A recombinant inbred population developed by single seed descent breeding technique from F2 to F7 generations was produced from a cross between BTx623 x PI568016 (Niu Sheng Zui), consisting of 292 inbred lines, evaluated for expression of early season cold tolerance based on field and controlled condition(l 5°C) germinability/emergence and seedling vigor.
