Purification of the G40001 accession was completed over several generations of single-seed descent in a screenhouse at the USDA-ARS-TARS in Mayaguez, Puerto Rico. This purified seed was then used for generating the genome sequence of G40001 (manuscript in preparation). The name "G40001-Seq" indicates that it is the genotype used for the genome sequence. The original G40001 accession was received from CIAT (International Center for Tropical Agriculture Research). In 2015, a single plant selection from this original seed lot, 15SH-6302, was used as the genetic stock for the Tepary Diversity Panel (TDP) and designated TDP-1. In 2016, a single plant selection from TDP-1 was completed, 16SH-9517-3, followed by another single plant selection, 17SH-1669-3 in 2017. Thus, a total of three cycles of single-seed descent were completed. Forty seed from 17SH-1669-3 were then planted in April 2017 in the Mayaguez screenhouse and harvested in bulk, and designated 17C4. Plants of 17C4 were dark adapted on two occasions in a dark, air-conditioned room for one day and then young trifoliolate leaf tissue was harvested and immediately frozen in liquid nitrogen, and subsequently stored at -80C for high molecular weight DNA extraction at the University of Arizona. The 17C4 seed lot was planted and further increased in the Mayaguez screenhouse in the Fall of 2019 to generate sufficient bulked seed for submission to the GRIN collection and for local storage.
